Research Abstract

ラット顆粒膜細胞培養を用いてゴナドトロピンレセプターの発現調節の検討を行ってきたので、今回も同様の培養細胞を使用した。
FSHレセプターに関して、cAMP-A キナーゼ系は転写のレベルで、合成促進に働くという結果を得てきたが,Cキナーゼ系の刺激が、一時的にmRNAの減少をもたらし、その後上昇に転じる反応も存在することを観察した。LHレセプターについてはCキナーゼ系によっては恒常的にmRNAレベルの減少があらわれた。この結果を考慮して、FSHおよびLHレセプター遺伝子とルシフェラーゼリポーターを含むプラスミッドに接合し、この細胞に導入後FSH,LHを作用させた時のルシフェラーゼ活性を測定した。
ルシフェラーゼ活性はこの培養系を用いて観察されたので、PRLのレセプター発現に対する影響を検討しているが、現在まで一定した結果を得ていない。しかし、PRLはLHレセプターのmRNAの有意の減少をもたらすことが観察された。
