| Research Abstract |
癌細胞の浸潤を制御している分子機構を解明し、positiveに働く経路を阻止することにより癌浸潤を制御することは理論的に可能である。この着想の元に本研究では、口腔扁平上皮癌ならびに唾液腺癌細胞を用いて浸潤機構を解析してきた。その結果、口腔癌の浸潤・転移に強く関与する分子は、デスモソームやEカドヘリンなどの細胞間接合分子、sialyl Lewis^a、さらに matrix metalloprotease(MMP)特に、MMP-2,MMP-9、膜型MMP(MT1-MMP)であった。in vitro浸潤モデルでの癌細胞の浸潤ならびにこれらのプロテアーゼの産生はEGFやTNFαなどのサイトカインによって促進されるが、これらの促進作用に転写因子(AP-1,NFkBなど)の活性化が強く関与していた。即ち、デキサメサゾンやプロテアソームインヒビターなどにより転写因子の活性化を抑制することにより、癌細胞の浸潤は著しく抑制された。異常の結果はMMP-9などのプロテアーゼは口腔癌の浸潤・転移抑制治療の標的分子になりうることを強く示唆している。
