| Research Abstract |
ビチセチンの全アミノ酸配列の決定:既にBitis arietannsの蛇毒より単離したビチセチンの物理化学的性質が、類似の活性を持つボトロセチンとかなり異なることが判明したので(Biochen Biophys.Res.Commun.(1996)226,273-279),その構成α鎖(131残基),β鎖(125残基)の全アミノ酸配列を決定した。ボトロセチンα,β鎖との相同性はそれぞれ47.1%,43.3%と低く,作用機序に相違のある可能性を示した(Res.Commun.Cell Mol. Biol.(1998)in press)。現在vWF分子上のビチセチン結合部位を探索中である。
ハリスターゼの精製、作用と全アミノ酸配列:Agkistrodon halys blomhoffi I(マムシ)の蛇毒よりセリンプロテアーゼ活性を持つ成分を精製し、ハリスターゼと命名し、アスパラギン(N)型糖鎖を含む1本鎖(238残基)から構成されているその全アミノ酸配列を決定するとともに、ヒト血漿タンパク質に対する作用を調べた。その結果、キニノーゲンに作用してブラジキニンを遊離すること、フィブリノーゲンを分解するがフィブリンを生成せず、むしろトロンビンによるフィブリン形成を阻害することなどが判明した(Eur.J.Biochem.(1998)in press)。
マムシギンの精製、性質、及びcDNAクローニングによる全アミノ酸配列の推定: マムシ蛇毒より血小板GPIbに結合し、血小板凝集を惹起する成分を精製し、マムシギンと命名した。cDNAライブラリーを作成し、構成α,β鎖のN末端アミノ酸配列をプローブとしてcDNAを単離、解析したところ、それぞれ21残基23残基のリーダーペプチドを持つ前駆体タンパク質として合成され、136残基,127残基の成熟タンパク質にプロセスされることが判明した。得られたアミノ酸配列は既知のGPIb結合タンパク質などと高度の相同性を示した(Thromb.Haemost.(1998)in press)。現在、大腸菌やバキュロウイルスによる発現実験を計画している。
