肝細胞増殖因子(HGF)の医薬への応用に向けての活性制御因子の生体内動態の解析

| Research Abstract |
肝細胞増殖因子(HGF)は強力な肝再生因子であり、現在肝疾患の治療薬としての開発が進められている。HGFの活性は細胞外、細胞内の種々の因子により制御されている。本研究ではそれらの因子についてHGF活性の制御との関係において解析し、以下の結果を得た。
1.肝障害時にHGFを活性化するセリンプロテアーゼとして同定されているHGF activator(HGFA)について、肝障害時における動態を解析したところ、HGFAのmRNA量が肝障害に伴って増加することが明らかになった。またHGFA自身の活性化も肝障害に伴って起こっていることが明らかになった。
2.HGFAの活性を阻害するインヒビターとして2種類のHGFA inhibitor(HAI)が精製されている。1型HAIについては、cDNAクローニングにより膜結合型として合成されプロセシングにより細胞外に分泌されると推定されている。1型HAIに対する抗体を作成し、イムノブロット法で1型HAIの動態を解析したところ、膜結合型1型HAIが同定され、また2段階のプロセシングにより、活性の高い1型HAIが細胞外に分泌されてくることが明らかになった。2型HAIについて、cDNAクローニングにより構造解析したところ、1型HAIと同様膜結合型として合成され、プロセシングにより細胞外に分泌されることが明らかになった。
3.細胞内においてHGFの活性を制御する新しいシグナル伝達分子として、HrsとHbpが同定されている。このHrs・Hbp経路と相互作用する分子を検索し、HbpのSH3ドメインに結合する分子としてUbiquitin C-terminal hydrolaseを同定した。この結果、Hrs・Hbpはこの酵素と相互作用することによりHGF/受容体複合体の細胞内分解に関与することが示唆された。
