| Research Abstract |
申請者らは、Rhシステムの遺伝子解析を進め、以下のような成績を得ることが出来た。
(1)LA-PCR法により増幅して得られたRHD遺伝子、RHCE遺伝子の全イントロン(各9個、計18個)のPCR産物と両遺伝子間のスペーサー領域のPCR産物をベクターに導入し、サブクローニング後、シークエンスを行い、約9割の領域について塩基配列を決定した。
(2)RHD遺伝子のプロモーター領域の解析を行い、すでに発表されていたRHCE遺伝子の同領域との比較検討を行った。
(3)RH遺伝子のイントロン解析中に見出されたイントロン8内に存在するshort tandem repeat(STR)について、種々のRh表現型を有する230名の日本人(RhD+:100名、RhD-:130名)の分析を行い、多型性を確認するとともに、RH遺伝子の分子進化を辿る有力な手掛かりを得ることができた。
(4)-D-、partial D、Rhnullなどの各種Rh変異型について、遺伝子解析を行い各々の変異部位を同定した。なお、partial DのDvaについては、種々の変異を見出し、遺伝子レベルでの分類を試みるとともに、遺伝子診断法を考案した。
