Research Abstract

現在までに、1)HGFが平滑筋細胞に作用を持たない内皮特異的増殖因子であること(J Hypertens 1996 ; 14 : 1067-1072 ; Hypertension 1996 ; 28 : 409-413)、2)HGF系が血管平滑筋、内皮及び心筋細胞に存在すること(Biochem Biophys Res Commun 1995 ; 215 : 483-488 ; Biochem Biophys Res Commun 1996 ; 220 : 539-545 ; Hypertension 1997 ; 30 : 1488-1454)、3)局所HGF系がアンジオテンシンIIやTGF-bによって負に調節され、プロスタグランディンによって正に調節されていること(Diabetes 1997 ; 46 : 138-142 ; Diabetologia 1997 ; 40 : 1053-1061 ; Hypertension 1997 ; 30 : 1448-1454)、4)高血糖性内皮傷害に局所HGFの産生低下が関係すること(Diabetes 1997 ; 46 : 138-142 ; Diabetologia 1997 ; 40 : 1053-1061)、5)虚血により血管壁HGF産生が低下し、内皮細胞のアポトーシスを誘導すること、6)腎局所HGF系も存在しており、メサンジウム細胞より分泌されるHGFが糸球体内内皮細胞や上皮細胞の増殖制御に関わっていること(Kidney Int 1998)、7)血清HGF濃度は高血圧患者で増加しており、血圧と正の相関を示し、合併症の診断に利用可能であること(Hypertension 1996 ; 28 : 409-413 ; Biochem Biophys Res Commun ; in press)、8)閉塞性動脈硬化疾患者で血管局所HGF産生低下と血清HGF濃度増加が認められ、PVDの重症度判定に血清HGF濃度測定が有用であること、などを報告した。これらの研究は、HGFが血管系においても極めて重要な因子であることを世界で初めて証明したものである。
