| Research Abstract |
Ribozymeは配列特異的に標的のRNAをmultiple-turn overで切断する活性があり、その抗HIV-1活性を検討した。私たちはHIVの細胞へのentryの際に重要な役割をするgp120のV3 1oopの中に標的を見つけRibozymeを合成してin vitroとin vivoでその抗ウイルス活性を検討した。Ribozymeを用いて遣伝子治療を行う際に細胞内のribonuclaseに対する安定性を高くするために安定化ribozyme (phosphorothioate DNA/RNAキメラRibozyme)を作成して実験に供した。標的としてはV3 1oopを含む346bpのDNA fragmentをPCRで増幅し、安定化rbozymeは野生化ribozymeよりも切断活性が若干低下するだけであることを確認した。また共焦点顕微鏡を用いて、安定化ribozymeは細胞内でも野性型ribozymeよりも、より安定であることを証明した。高感度な感染系を樹立するためにluciferase遺伝子をもち、ribozymeが作用しないようにV3領域を欠落させた、リポーター遺伝子を用いた。これらの遺伝子と様々なenv遺伝子を細胞に共導入することにより偽ウイルスを作製して、感染実験に供した。合成したRibozymesはenvelope介する細胞融合を抑制し,NL432とSF162ウイルスの感染も効率よく阻止した。また、ribozyme処理細胞ではWestren blotでenv蛋白の発現が低下したので、感染阻止はribozymeによるenv発現の特異的抑制効果によると考えられた。これらのribozymeによる感染予防も視野に入れ応用をめざしたい。
