| Research Abstract |
Genetic population structure of the commercially fished loliginid squid Photololigo edulis was examined by collecting three samples (294 individuals) of two seasonal populations from the East China Sea, and subjecting individuals to electrophoretic analysis.
Twenty-three enzymes were tested, and 10 enzymes were screened for polymorphism. Clearly resolved enzyme phenotypes were obtained at seven (malate dehydrogenase, MDH-1ィイD1*ィエD1, MDH-2ィイD1*ィエD1, MDH-3ィイD1*ィエD1, E.C. 1.1.1.37 ; phosphogluconate dehydrogenase, PGDHィイD1*ィエD1, E.C.1.1.1.44 ; aspartate aminotransferase, AAT-2ィイD1*ィエD1, E.C.2.6.1.1; adenylate kinase, AK-2ィイD1*ィエD1, E.C.2.7.4.3 ; phosphoglucomutase, PGMィイD1*ィエD1, E.C.5.4.2.2). Routine scoring at the 7 loci revealed low levels of genetic variability, with the proportion of polymorphic loci, P=0-0.1, mean heterozygosities per locus=0.28-2.19 x10ィイD1-2ィエD1.
Analyses of allele frequencies at seven polymorphic loci provided no evidence of genetic separation, and the frequency distribution of genotypic classes almost fitted Hardy-Weinberg expectations for randomly mating population. Nei's mean genetic distances between samples ranged from 0.491-2.522(x10ィイD1-4ィエD1), respectively, supporting the opinion that genetic population structure of the squid is homogeneous.
