Research Abstract |
A series of experiments were conducted to evaluate phytin phosphohydrolysis activity in the rumen and to isolate phytase positive rumen bacteria. Bacterial phytase activity was suggested not to be high. None of the phytase positive bacteria was isolated under anaerobic condition irrespective of the medium used. Two bacterial colonies showing clear zones were seen on PSM plates incubated under aerobic condition. One bacterium exhibited pinkish opaque colonies that made larger halo and the other yellowish colonies making relatively small halo. The numbers of phytase-positive bacteria in the rumen increased from nearly undetectable level to 1.7x10ィイD15ィエD1 cfu/ml 1.0h after morning feeding, thereafter declined to the initial levels at 7.0h after morning feeding. Kpu 001 was Gram negative non-motile straight rod and able to grow both under anaerobic and aerobic condition. According to the physiological and morphological results suggested Kpu 001 to belong the genus Klebsiella and most likely to be K.pneumoniae. Kpu 002 was suggested to belong to the genus Corynebactreium Phylogenetic analyses on 16S rDNA indicated that Kpu 001 (DDBJ Accession No : AB012208) was closely related to Klebsiella pneumoniae JCM 1662 and Kpu 002 (DDBJ Accession No. : AB012207 was to Corynebacterium vitarumen. The presence of potent phytase producing microorganisms in the rumen was questioned.
