Research Abstract

1.サイクリンE発現ベクターの作成
(1)米国スクリップス研究所S.Reed博士より供与されたサイクリンEcDNAの全コーディング領域を含むEcoRI断片(2.5kbp)をサイトメガロウイルスプロモーターを有する真核生物発現ベクターpCDNA3にサブクローニング後,制限酵素断片解析ならびにインサートのシークエンシングにより,恒常的サイクリンE過剰発現ベクターを得た.
(2)(1)により得られた恒常的サイクリンE過剰発現ベクターを,リポフェクチン法によりNIH/3T3マウス線維芽細胞株に導入した.導入後,ネオマイシン誘導体G418にて選択を行い,pCDNA3のみの導入株を含め,各細胞株につき10クローンを回収した.
2.サイクリンEアンチセンスベクターの作成・導入
(1)1-(1)で使用したサイクリンEcDNAの転写開始部位を含む110bp断片をpCDNA3にアンチセンス方向にサブクローニング後,制限酵素断片解析ならびにインサートのシークエンシングにより,サイクリンEアンチセンスベクターを得た.
(2)(1)により得られたサイクリンEアンチセンスベクターを,リポフェクチン法によりサイクリンE過剰発現株TMK-1,MKN-7胃癌株,サイクリンE低発現株MKN-28,MKN-74胃癌株に導入した.導入後,ネオマイシン誘導体G418にて選択を行い,ベクターのみの導入株を含め,各細胞株につき10クローンを回収した.
