Research Abstract

本研究の目的は、PTCA後ヒト冠動脈の新生内膜形成における平滑筋細胞、マクロファージ、および内皮細胞の動態と、それに関与する種々の血管作動性物質のリガンド-レセプター系、特に代表的な血管収縮物質であるアンジオテンシンIIとアンジオテンシンIIタイプ1レセプター系、同じくエンドセリンとエンドセリンAレセプター系、および血管拡張物質であるナトリウム利尿ペプチドとそのレセプター系(GC-A,GC-Bおよびクリアランスレセプター)のup-regulationの有無と遺伝子発現について、免疫細胞化学的方法、in situ hybridization、およびRT-PCR法を用いて分子病理学的に解析し、ヒトPTCA後再狭窄における収縮性リモデリングの分子病理学的機序を解明することである。
本年度は、ヒトPTCA後剖検例の冠動脈凍結材料およびPTCA後再狭窄のアテレクトミ-材料を用いて検索し、新生内膜形成過程の平滑筋細胞の再分化機序ではミオシン重鎖アイソフォームの発現変換が起こることを初めて明らかにした(Circulation,96:82-90,1997)。その他、PTCA後新生内膜におけるアンジオテンシン変換酵素の発現亢進(Circulation,96:3328-3337,1997.J.Hypertens.,15:1295-1302,1997)およびエンドセリン変換酵素とエンドセリンの発現亢進(Circulation,96:I-348,1997)についても初めて明らかにした。
