Research Abstract	本研究の目的は、PTCA後ヒト冠動脈の新生内膜形成における平滑筋細胞、マクロファージ、および内皮細胞の動態と、それに関与する種々の血管作動性物質のリガンドーレセプター系、特に代表的な血管収縮物質であるアンジオテンシンIIとアンジオテンシンIIタイプlレセプター系、同じくエンドセリンとエンドセリンAレセプター系、および血管拡張物質であるナトリウム利尿ペプチドとそのレセプター系(GC-A,GC-Bおよびクリアランスレセプター)のup-regulationの有無と遺伝子発現について、免疫細胞化学的方法、in situ hybridization、およびRT-PCR法を用いて分子病理学的に解析し、ヒトPTCA後再狭窄における収縮性リモデリングの分子病理学的機序を解明することである。我々は本研究において、ヒトPTCA後の新生内膜形成過程における平滑筋細胞の再分化機序では、ミオシン重鎖アイソフォームの発現変換が起こることを初めて明らかにした(Circulation,96:82-90,1997)。その他、PTCA後新生内膜におけるアンジオテンシン変換酵素の発現亢進(Circulation,96:3328-3337,1997.J.Hypertens.,15:1295-1302,1997)およびエンドセリン変換酵素とエンドセリンの発現亢進(Circulation,96:I-348,1997.)についても初めて明らかにした。さらに、ヒト冠動脈新生内膜におけるエンドセリン A レセプターの発現亢進についても報告した。(Circulation,98:I-734,1998)
