Research Abstract |
We searched and identified for new members of glycosyltransferases involved in the synthesis of Lewis antigens which are the ligands for selectins. Successfully, we have cloned two novel genes encoding new glycosyltransferases, and named Fuc-TIX and beta3Gal-T5. Fuc-TIX was mainly expressed in brain and kidney, and participated in the synthesis of CD15 (Le^x) antigens. beta3Gal-T5 was found to be responsible for the synthesis of type 1 Lewis antigens, such as Le^a, Le^b and sLe^a. In particular, B3GaI-T5 was proven to be a responsible enzyme for the synthesis of CA 19-9 (sLe^a) antigen in gastrointestinal tissues. CA 19-9 is a very famous tumor marker, and its carbohydrate structure is defined as sLe^a which is the selectin ligand.
