Research Abstract	ジーントラップに用いるトラップベクターはEn-2イントロンの下流にスプライシングの受容シグナル、翻訳活性を上げるためのIRESシグナル、およびLacZをマーカー遺伝子として配置、下流にはアクチンプロモーター下にネオマイシン耐性遺伝子を挿入し、その有効性は確認済みである。このベクターをES細胞株であるD3およひE14-1細胞に導入して数千個のES細胞クローンを樹立した。これらのクローンをプールしてop/opマウス由来の骨髄繊維芽細胞株OP-9上で培養し、8日目にX-gal染色を行う方法でスクリーニングした。LacZを発現する陽性細胞は非常に少なく同定は困難であったが、陽性と思われる細胞を選別後、cDNAライブラリーを作成し、クローンを同定した。10種類あまりのcDNAの塩基配列を決定後、未知と思われるものをプローブにして成熟マウスの各組織での発現をノザン法にて解析した。多くはubiquitousな発現が見られたが、クローンによりある程度の組織特異性はみられるものの、造血系組織で特異的に発現しているクローンは得られなかった。 ES細胞はin vitroで種々な組織・細胞に分化しうるがheterogenousであり、ジーントラップで血球系細胞特異的な発現が見られるクローンを同定するには特定の系列の細胞に分化する系を確立する、あるいはFACSを用いて特定のlineageの細胞をソートしてくる必要があると考えられ、現在も両方面からの系の確立を試みている。将来的にはマウス以外のES細胞を用いることも視野に入れて研究を続ける。
