Research Abstract

It is well-known that Lewis phenotypes change from birth into early childhood. To begin to elucidate the cause of the change, we determined Lewis phenotypes and Lewis (Le) and secretor (Se) genotypes in samples of cord blood and infants blood by PCR-RFLP.We also quantitated the Lewis antigens in bloodstains and plasma of those samples by time-resolved fluoroimmunoassay. The Lewis phenotypes of cord blood (71 cases) and 21 newborn infants' blood (6 days) were Le(a-b-). At 1 month after birth, Le(a+b-) began to be observed and Le(a+b+) was observed from 2 months to 10 months of age. Le(a+b+) phenotype would subsequently change to Le(a-b+) by Le and Se genotyping (Le/-, Se/-). It was found that Le(a-b+) phenotype began to appear from about 9-months. These phenotype was genuine. Le^a antigen and small amount of Le^b antigen were detected in cord blood
In se/se genotype, the amount of these antigens in plasma of cord blood were significantly high. The Le^b antigen began to increase from 5 days after birth. It is suggested that the antigen is adsorbed to blood cell after birth. The components of plasma of cord blood and adult blood were analyzed by polyacrylamide gradient gel. Plasma proteins less than molecular weight 24,000 were found in cord blood, but not found in adults' blood. The concentrations of high density lipoprotein (HDL) and low density lipoprotein (LDL) were lower in cord plasma than those in adult plasma. Three LDL components were detected in cord blood, but the highest molecular weight component among them was detected in adult plasma, From these results, we suspected that these low molecular weight proteins were a carrier of Lewis antigen in cord blood and these induced the difference of Lewis antigen values on red blood membrane.
