| Research Abstract |
本研究はヒト自己免疫疾患のプロトタイプとされる全身性エリテマトーデス(SLE)の病因解明のため、自己抗原のクリプティクエピトープに対する活性化T細胞が存在するか否かおよび自己抗原クリプティクエピトープ反応性T細胞が実際に生体内でクローナルに増殖しているか否かを検討する。最近、我々はTCRβ鎖遺伝子の配列解析から、SLE患者末梢血中にはオリゴクローナルなT細胞の増殖があることを見いだしたので(Int.Immunol.)、対象自己抗原を52kD蛋白やカルレティクリンなどのSS-A関連蛋白に絞らす、上記SLE患者の生体内で増殖しているT細胞の認識する抗原全般について検討することとした。そのために、今年度行った増殖T細胞クローンの解析ではSLEの急性増悪の前後で増殖T細胞のクロノタイプが変わってしまうこと(論文作成中)、および急性増悪時にはCD4陽性T細胞のクローナルな増殖が著しいこと(論文作成中)を発見した。この急性増悪時に出現するT細胞の対応抗原を広く検索する目的で、当該T細胞のTCRを再構築を目指した。具体的には単一細胞からT細胞受容体遺伝子を増幅する方法を確立した。現在、この方法を用い、当該T細胞のT細胞受容体を蛋白レベルで再構築することを試みている。来年度はこれを用いて対応自己抗原を同定し、クリプティクエピトープに対する反応性を含め、自己抗原T細胞エピトープを検討する予定である。
