Research Abstract	これまで全身性エリテマトーデスの免疫学的異常に関わる因子のひとつとしてT細胞の異常な感作が報告されている。また、自己抗原に対するT細胞の反応が観察されたり、マウスでは自己抗原のクリプティックなエピトープが自己反応性T細胞の活性化に関与しているなどの報告がある。これまで、我々は患者において末梢血中にオリゴクローナルなT細胞の増殖があることを報告しているが、これらのT細胞の対応抗原は明らかでない。本研究ではクリプティックなエピトープも含め、全身性エリテマトーデス急性増悪時に集積するT細胞のエピトープ(抗原ペプチド)を同定するために1個のT細胞からT細胞受容体のα鎖およびβ鎖遺伝子をクローニングする方法を開発した。これにより、患者体内に実際に集積しているT細胞のT細胞受容体が容易に再構築されると考えられる。また、MHCクラスIとともに抗原ペプチドがファージ上に発現されたライブラリーの作成をめざし、まず、実際にMHCクラスI分子遺伝子をβ2-マイクログロブリン遺伝子に接続し、一本鎖蛋白としてファージ上に発現されうることを確認した。
