| Research Abstract |
C型肝細胞癌患者血清中からC型肝炎ウイルス(hepatitis C virus:HCV)RNAを抽出し,reverse transcription-PCR法にて各HCV遺伝子,その中でもまずインターフェロン治療効果を規定しているといわれるNS5A遺伝子の一部分を,酵母内でGAL4 DNA binding domain(BD)との融合蛋白を発現するplasmidにcloningした.そのplasmidを用いて,GAL4 UASの下流にbeta-galactosidase遺伝子が組み込まれている酵母をtransformしたところ,X-gal染色によりplasmidの導入されたすべての酵母が青くなることが確認され,酵母内でNS5A蛋白の一部が転写活性化因子として機能する可能性が示された.NS5A蛋白のHCV感染肝細胞内での機能を検討するため,様々なNS5Aのdeletion mutantとGAL4 DNA BDとの融合蛋白を哺乳細胞内で発現するplasmidを構築し,その転写活性化能につき検討した.全長のNS5A蛋白には転写活性化能は認められなかったが,N端を欠失したNS5A蛋白が肝癌細胞を含む哺乳細胞内で転写活性化因子として機能することを見出し,またその転写活性化domainを決定した.転写活性化domainには転写活性化因子の共通domainである酸性領域を2カ所,プロリンに富んだ領域を1カ所認めた.また,N端を欠失したNS5A蛋白に転写活性化能があることから,NS5A蛋白とヒトの基本転写因子群との相互作用につきGST pull-down法およびimmuno-precipitationにて検討し,TATA binding protein(TBP)およびTBP associated factor(TAF)の一部がNS5A蛋白と結合することを認めつつある.
