炎症性腸疾患・遺伝子欠損マウスの虫垂リンパ装置と腸内抗原に関する免疫学的研究

Research Abstract

1) 大腸内視鏡検査時に採取した虫垂/腸管の粘膜リンパ装置,単離リンパ球についてそれぞれ免疫組織学的及びフローサイトメトリーにて検討し,32例の臨床例で健常人との差違を認めた。すなわち、潰瘍性大腸炎患者(n=32)の非病変部虫垂リンパ球は健常人の27%に比しCD4陽性αβT細胞が有意に増加していた。一方、直腸病変部ではγδT細胞の有意な減少(健常群:5.2%、潰瘍性大腸炎:1.9%)とCD4陽性細胞の増加(健常群:16%、潰瘍性大腸炎群:25%)を認めた。
2) Balb/cおよびC/BL6近交系の胸腺摘出マウスの妊娠12-15日目にIL-7受容体抗体を投与し小腸パイエル板欠損マウスを作成。パイエル板欠損マウスと虫垂切除を加えたマウスにデキストラン硫酸(DDS)で腸炎を惹起させるとパイエル板欠損マウスでは有意な大腸炎の発症を抑制した。KLH経口投与マウスでは、虫垂切除によりTh2/Th1型抗体価比の上昇を認め、虫垂粘膜リンパ装置と全身免疫との関連性を認めた。
