Research Abstract |
We tried to induce a T cell liesponse to hepatitic Cvirus (HCV) core protein by using a DNA vaccine in mice. We constructed plasmids encoding HCV core region under the control of various promotors such as pMAMneo, pEFBOS, and pcDNA3. These plasmids alone induced a helper T cell response only after in vitro restimulation twice. A cytotoxic Tlymphocyte (CTL) response was observed but the magnitude was not strong. Next we immunized mice with a plasmid encoding HCV core region together with an adjuvant plasmid encoding IL-2, GM-CSF, or (CD86. None of these combination resulted in a betterT cell response yet.
