| Research Abstract |
我々は、昨年度まで、HCVの新たな予防、治療法の開発のため、DNAワクチンに取り組んできた。今年度は、新たな治療法の開発のためHIV感染症に対して取り組まれているAntisense RNAの基礎的研究をおこなった。Antisense RNA constructの開発のため、5'-noncoding regionとcore regionを含む7つのAntisense RNA constructを作製し、in vitro translation systemを用いてHCV core蛋白の発現抑制効果に関して検討した。その結果、5'-noncoding region全領域とcore領域の開始コドンから約60bp(nt 1-nt 402)を含むAntisense RNA constructにおいて、約50%のHCV core蛋白の発現阻害効果が認められた。さらに、このAntisense RNA constructとHCV-luciferase発現constructを、 HuH7細胞へのco-transfectionして、luciferase活性にてAntisense RNAの抑制効果を調べた。その結果肝癌細胞内でも同様に約50%程度のAntisense RNAの抑制効果が認められた。今後は、このAntisense RNAの細胞内導入法と効果増強法に関してさらに検討していく予定である。
