| Research Abstract |
我々はidetical twinとそのmother、elder brotherにてtax mutation、HTLV-I proviral DNA load、抗HTLV-I抗体価、リンパ球幼若化反応、HLAタイピングをそれぞれ検討した。
1) 検体DNA中のHTLV-Iプロウイルスのtaxコード領域をnested-PCR法にて増幅、20クローンをランダムに選び、塩基配列の決定を行った。塩基配列の解析はプロトタイプλATK-1と比較した。Identical twinにおける各クローンの塩基配列の解析結果では、HAM発症のtwinはtax領域に変異のみられたクローン数は18クローン中8個、carrierでは18クローン中18個であった。アミノ酸置換を伴う変異はHAM3個、37.5%、carrierで9個、52.9%であった。
2) HTLV-I proviral DNA load定量を行ったが、proviral DNA量はHAMで多い傾向を認めた。
3) 抗HTLV-I抗体価は従来の報告どおりHAMにおいて多い傾向を認めた。
4) リンパ球幼若化反応はHAMにおいてspontaneous proliferationの亢進を認めた。
5) HLAタイピングの結果はmother,identical twinの3者に共通して従来HAM型といわれているハプロタイプがみられたものの、motherではCw3DR9、identical twinではDR9DQ3などATL型も認められた。
6) ENV領域の塩基配列を決定しHLAのエピトープである可能性がある変異をキャリヤーに認めた。
7) tax領域では5箇所の変異が認められ、1箇所のHLAアンカーモチーフとなり得る変異を見いだした。
