Research Abstract |
It was made that this study produced the antibody for the human IGF-II receptor and that it makes the assay system to be a goal. Results are as follows. 1. The purification from the placenta of the human IGF-II receptor : Though it succeeded, the yield point is 3.3 μg/g tissue. 2. Expression and purification in a Escherichia coli of the human IGF-II receptor : The extracellular part of the IGF-II receptor was divided into five, each cDNA was included in the expression vector. Though the expression was confirmed on third part ( the 951-1384 residue ) and fifth part ( the 1821-2304 residue ), the confirmation was not possible on the other part. 3. Peptide production of antibody for the human IGF-II receptor : 3 kinds of peptides of the human IGF-II receptor of 7-22 residue, 1103-1119 residue, 2046-2064 residue were chosen as an antigen. The carrier protein was combined, and the antibody for all peptides was got. At present, IgG fraction of the rabbit serum is being refined. The construction of assay system of the human IGF-II receptor is tried after the purification. The development of sandwich process and competition method which utilizes the mini IGF-II receptor as a standard is considered. It is parallel to the development of the assay system, in the element of the informed consent, blood and liquor amnii of the pregnant woman, sampling of IUGR children, low statue infants, blood of the overgrowth child, etc. besides have been started.
