Research Abstract

CD36 is one of the major receptor for oxidized low density lipoproteins (LDL).We have found that CD36 is expressed in macrophages resided in atherosclerotic lesions and its distribution in the lesions are different from that of scavenger receptor class A, in that CD36 is expressed dominantly in lipid-rich macrophages.To know the physiological function in vivo, we have tried to create the mice lacking this receptor in this research project.We have screened the C129 mouse genomic library with using murine CD36 cDNA as a probe and successfully cloned the genome encoding CD36.Based upon the restriction map of CD36 we made, we have constructed the replacement type targeting vector which lacks exon 1 containing the translation initiation codon of CD36.After electroporation of the construct DNA into embryonic stem (ES) cells, we have screened them with Neomycin and obtained five clones with homologous recombination, determined by Southern blot analysis.We have injected the positive ES clones into blastcysts and waiting for the chimeric mice's being born.
