Research Abstract |
To measure NO concentration in muscle, we improved near-infrared spectroscopy and its computer analyzing system. We saturated hemoglobin medium with NO in several defined concentration, and measured NO hemoglobin absorbance as reference. In this modification, we could measure NO concentration in vitro. In the experimental model, however, using New Zealand White rabbits, it was impossible to measure NO concentration in the muscle. But it was possible to assess the recovery of the oxygenation in the calf muscle after hindlimb ischemia caused by the ligation of the femoral artery. Therefore, we investigated that recovery process in the ischemic limbs in the rabbit experimental model. The rabbits with ischemic limbs were divided into four groups. Group A ; which were fed on a standard diet, Group B ; which were fed on a high-cholesterol diet, Group C ; which were fed on a standard diet and treated with 5HT receptor antagonist (anti-platelet agent), Group D ; which were fed on a high-cholesterol diet and treated with 5HT receptor antagonist. There was no difference between Group A and Group C in ischemic recovery time (T1/2). T1/2 in Group D was shorter than that in Group B.There was statistically difference between Group B and Group D in T1/2. T1/2 in Group B was statistically longer than that in Group A.5HT of Group B in serum was higher than that in Group A.On the other hand, 5HT of Group B in whole blood was lower than that in Group A. These data showed that 5HT receptor antagonist improved the muscle oxygenation after ischemia in this model. This study demonstrated that it was possible to measure tissue oxygenation in the animal model during exercise, as well as in the patients with atherosclerosis obliterans. The near-infrared spectroscopy analysis provide the valuable information to assess the drug effect not only in clinical setting but also in experimental model.
