Research Abstract |
Loss of heterozygosity (LOH) was analyzed in breast cancer using 40 microsatellite markers on over 20 chromosomes. LOH was frequently seen in 17p (44%), 16q (31%), 17q (29%) and 13q (23%) in invasive ductal carcinama of the breast. Accumulation of LOH in individual patient was well associated with high histological grade and a significantly poor prognosis. This shows that accumulation of LOH occurs at a late stage of breast cancer progression and may be a useful prognostic marker of breast cancer. Furthermore, LOH in 16q was also frequently seen in ductal carcinoma in situ (DCIS). This shows that this chromosomal region may be important in early stage of carcinogenesis in DCIS. We investigated on the mRNA expression of ATBF1 gene located in 16q22.3-23.1 using RT-PCR method in breast cancer cell lines (MCF7, T47D, ZR75- 1, BT-20, MDAMB231, SKBR3) and clinical breast cancer cases. However, the level of mRNA expression was quite low in breast cancer and normal breast tissues. The expression was highly seen in normal lung and bronchial tissues, especially normal tissues. In situ hybridization showed that normal bronchial cell expressed high levels of ATBF1 mRNA signals. These data shows that AIBF1 gene may play an important role of genesis and growth of normal lung tissues.
