Research Abstract

1. The changes of cytokines in the liver tissue after pancreatectomy - The effect of administration of mitomicin C into portal vein
Under intraperitoneal pentobarbital anesthesia, male balb/c mice were given distal pancreatectomy and splenectomy. The animals in the MMC group were administered mitomycin C (1mg/kg) into the portal vein soon after pancreatectomy, while the animals in the control group recieved no medication. The animals were killed 0 to 24 hours after surgery, and liver tissues were excised. After homogenization of the liver and centrifugation, tumor necrosis factor alpha, jnterleukin (IL) 1 beta , and IL 6 levels were determined by an enzyme-linked immunosorbent assay.
Tumor necrosis factor alpha and IL I beta did not change throughout the experiments in either groups ; however, IL 6 in the liver became significantly elevated from 0.680.27 and 0.43*0.20 pg/mg protein to 11.5*3.2 and 8.17*0.60 pg/mg protein, respevtively in the control and MMC groups, 30 minutes after surgery. This significant elevation of IL 6 persisted for 24 hours and 12 hours after surgery in the control and MMC groups, respectively.
2. Expression of the E-selectin (ELAM-1) gene in the liver tissue after pancreatectomy
E-selectin gene expressions in the liver were assayed by a reverse transcriptase-nested polymerase chain reaction. Expression of the E-selectin gene in the liver was not detected throughout the experimental period, while expression of the GAPDH gene was detected in every samples.
Thus, we concluded that pancreatectomy promoted IL 6 production in the liver and that rnitomycin C injection into the portal vein might reduce this elevation.
