Research Abstract

平成9年度は膵臓癌細胞が肝転移を形成する際に発現が促進あるいは抑制される遺伝子群を同定するため膵癌細胞株における肝転移群と非肝転移群各群の間でDifferential DisplayおよびDNA sequencingを施行した。
(対象と方法)ヒト膵癌細胞株BxPc-3,SW1990,Capan-2,PaCa-2よりRNAを抽出しcDNAを作成、α-^<32>PdCTP、randam primerを用いDifferential Displayを施行した。
(結果)両群間に40個のDNA flagmentについて差を認めた。その塩基配列および発現の差異について現在同定中である。
現在までに、非転移群よりETS-2 3'end,KIAA00196,human STS WI-8550,E.coli dipeptidyl carboxypeptidase homologue,C.ekegance cosmid R01E6 homologue,human HTGS phase 1 clone 151B14 homologue
転移群よりBRCA2gene region chromosome 13q12-13 containing CA repeat,M.musculus gene for betaenolase homologue、γ-interferon homologue、C.elegance cosmid D1025 homologue、human STS WI8085等の一部に相同性があり、候補としてあげられ、現在、両群間の発現の差異をRT-PCR、Nothern hybridizationにて検討し、転移との関連性について精査中である。
