| Research Abstract |
脳腫瘍におけるテロメラーゼの発現を調べ、その診断的意義と治療への応用の可能性について検討した。まずTRAP法を確立し手術摘出標本あるいは培養細胞より蛋白質を抽出し、TSプライマー、CXプライマーを作用させPCRにて増幅しオートラジオグラフィーにてテロメラーゼ活性の有無を検索し、悪性化に伴いテロメラーゼ活性は上昇した。
in vitro実験による検討として、ヒトグリオーマ培養細胞を用い、これを分化誘導した場合のテロメラーゼ活性の変化を検討し、細胞の分化とテロメラーゼ活性の関連について検討した。その結果、cAMPによりグリオーマ細胞にて分化誘導されるとテロメラーゼ活性は著しい減少を示した。さらにこの変化に伴い増殖能は減少する傾向を示した。現在、in vivo実験による検討として、ラット脳腫瘍モデルを作り、前腫瘍病変、腫瘍病変におけるテロメラーゼ活性の変化を調べ、酵素活性の推移と、腫瘍発現時期の相関に関して検討中である。
