Research Abstract |
Recently a study of glutamic acid containing neurons in the brain have been carried out with the use of glutaminase, a synthetic enzyme of glutamic acid. This study was undertaken to identify the glutamatergic neuronal cell bodies and their terminals in the trigeminal motor nucleus and the trigeminal sensory nuclei, with the use of polyclonal antibody against glutamic acid. Female Wistar rats were used for this study. They were anesthetized deeply and were perfused with 2.0% paraformaldehyde and 0.25% glutaraldehyde. The brain stem was removed immediately and serial frozen sections were made. Immunohistochemical labeling of glutamate - immunoreactive neuronal cell bodies and varicosities were performed with fluorescein isothiocyanate (FITC), and were observed under a fluorescent microscope. Glutamate-immunoreactive (Glu-IR) cell bodies and varicosities were detected by immunofluorescence technique. In the confine of cytoarchitechtonicaliy defined trigeminal motor nucleus (Vm), a few Glu-IR varicosities were observed on motoneurons and their proximal dendrites. Dense Glu-IR fibers were seen in the caudal subnucleus (Vc) of the spinal trigeminal nucleus (Vsp), and a number of Glu-IR celis were encountered in the trigminal main sensory nucleus (Vp) and the interpolar subnucleus (Vi) of Vsp. A number of Glu-IR cells were also observed in the parvocellular reticular nucleus (pcRN) adjacent to Vsp and around Vm.
