Research Abstract |
We established a tongue squamous cell cancer cell line, SCC-HOSO, which had been used for reseach of bone and calcium metabolism.In this study we have investigated the relation between calcium metabolism and apoptosis. (1) We examined the expression of oncogene and protooncogene in SCC-HOSO cell.We could detect the mutation of p53, although c-myc was not expressed in SCC-HOSO cell. (2) SCC-HOSO cell produces high level of parathyroid hormon related protein (PTHrP).Prostaglandin E2 increased the production of PTHrP of SCC-HOSO. In 4 subtypes of prostaglandin E2 receptors, SCC-HOSO expressed mRNA of EP2 receptor but not of the others.Fathermore, since prostaglandin E2 stimulated the intracellular cAMP of SCC-HOSO in vitro, EP2 protein was proved to be tanslated in this cell line. (3) Apoptosis of SCC-HOSO was not affected by either prostaglandin E2 or PTHrP.These agents were not stimulated the apoptosis of SQIS and SQFK, which were also cell lines of tongue squamous cell cancer.
