Research Abstract

1.bfp gene targetingキメラマウスの作製
上記のSouthern blot解析でpositiveであったES cellは、3.5日目のC57BL/6マウスの胚盤胞にマイクロインジェクションされ、偽妊娠したICRマウスの子宮上部に戻された。18.5日後にキメラマウスが生まれ、その毛色によりgermline transmissionの有無が確認された。
2.bfp gene targeting mouseの作製
上記の様にgermline transmissionが確認されたFIマウスは、5割の確立で変異アレルを持つヘテロ接合体マウスである。Southern blot解析にてヘテロ接合体マウスを同定し、そしてヘテロ接合体マウス同士の交配によりホモ接合体マウスも得ることが出来た。これらのマウスの脳、神経系を中心に解剖学的な構造解析や、行動解析を進行中である。
