The small GTP-binding proteins, including Rho, are implicated in the signaling cascade which regulates actin cytoskeleton-dependent cell functions. In this study, we have studied this small GTP-binding protein cascade at a molecular level. Regulation of activation of PHO1 : We have isolated and analyzed proteins which interact with ROM1/ROM2, GDPIGTP exchange factors for RHO1. Modes of action of RHO1 : We have isolated new proteins, SPA2 and SHS1, which bind BNI1, a potential target for PHO1. SPA2 is required for the localization of RHO1, and SHS1 is involved in cyotokinesis. Modes of action of BNR1 : We have isolated an SH3 domain-containing HOF1, as a protein which bind BNI1 related protein, BNR1. Both HOF1 and BNP1 are localized at the cell division site and involved in cytokinesis. As described above, we have analyzed PHO1-mediated small GTP-binding protein cascade and obtained new findings in the cascade at a molecular level.
