| Research Abstract |
膵臓がんの発生・進展に関与する遺伝子異常の同定を目標として、染色体領域のコピー数の異常をComparative genomic hybridization, fluorescence in situ hybridization,マイクロサテライト解析によって詳細に解析し、第6番染色体長腕の独立した3箇所、すなわち、6q21, 6q23-q24, 6q26に高頻度の共通欠失領域を同定した。これら共通欠失領域のうち69%の症例で欠失が認められた6q21の1-cMの領域をクローニングするために、YAC(酵母人工染色体)、BAC(細菌人工染色体),YACからサブクローニングしたcosmidを用いたコンティグを構築した。その結果、共通欠失領域は500-kbのYACにより完全にカバーされることを明らかにした。同時に、同領域の制限酵素地図を作成し、幾つかの箇所にCpG islandの候補領域を見出した。候補がん抑制遺伝子の単離のためにESTスクリーニング、cDNAセレクションを行い、複数の候補遺伝子を得て解析している。
