Research Abstract

ヒトLECT2レセプターの検出系の確立及びLECT2レセプターを発現する培養細胞のスクリーニングを目的として、CHO細胞由来のヒト組み換え体LECT2(rLECT2)をペルオキシダーゼ法により常法にしたがって125Iラベルし、各種細胞に対して結合実験を行った。用いた細胞は、接着系の培養細胞としてHeLa細胞,HepG2,HuH-6を初めとする肝癌細胞6種、及びMC3T3-E1細胞、血球系癌細胞としてHL-60,U937,K562など15種類の細胞をPHA(5μg/ml),LPS(10μg/ml),PMA(1ng/ml)で24時間処理しBinding assayに供した。接着系細胞でのassayは、10cmのdish当たり1×10^6まいた細胞に対して1,2,4nMの標識LECT2を処理し、4℃で3時間インキュベーション後、細胞と上清の放射活性を測定した。また、特異的結合については標識LECT2添加1時間後に非標識LECT2を8μM加え、さらに2時間インキュベーションした。浮遊系細胞では、1サンプル当たり5×10^6細胞に対しtest tubeを使い同様の実験を行った。スクリーニングの結果、ヒト血球系細胞HuT-78,K562,マウス骨芽球系細胞MC-3T3-E1が候補として得られた。しかし、cDNAのクローニングの為のLECT2レセプターmRNAのsourceとしては発現レベルが低いことが予想されたため、現在、さらに別の細胞のスクリーニング、高感度Binding assay系の検討を行っている。
