Research Abstract	我々はHHV-8の病原性、特に発癌との関連性を検討する目的で、HHV-8特異的単クローン抗体、HHV-8cDNAライブラリーの作製を行い、種々のウイルス遺伝子の解析を試みた。その結果、HHV-8特異的単クローン抗体B291は約50kDaのHHV-8特異的蛋白と反応し、ORF K9がコードするvIRFを認識することが判明した。その蛋白発現時期及び局在を共焦点レーザー顕微鏡にて検討したところ、薬剤未処理のBCBL-1細胞(ウイルス潜伏感染時期)ではvIRF発現が認められず、TPA処理(ウイルス増殖時期)で細胞質や核内に蛋白発現が認められた。しかしBCBL-1細胞をPFA+TPA処理(ウイルス複製の前初期)するとその発現は部分的に抑制された。一方、vIRF mRNAをRT-PCR法にて解析したところ、薬剤未処理BCBL-1細胞ではVIRF mRNAは検出されず、TPA処理にて発現が認められ、PFA+TPA処理にてその発現量が増すことも判明した。また、vIRF mRNAの構造は全長約1.5kbで、Kozak ATGやpoly A signalを有し、ゲノムからの転写に際しspliceされないことが確認された。以上の結果より、vIRFはウイルス増殖時期に発現するearlyまたはearly-late蛋白の一つで、細胞質、核内に局在することが明らかとなった。現在、この単クローン抗体B291を用いてvIRFの各種病変部位での発現の有無、生物学的機能解析などを行い、その病因論を検討している。
