| Research Abstract |
(1)ヒト胎児皮膚におけるcornified cell envelope構成蛋白の発現様式の免疫組織化学法、蛍光抗体法による観察
胎生8週から24週の各週齢のヒト胎児皮膚材料より、凍結切片を作成、抗ヒトloricrin small proline-rich proteins、involucrin抗体をそれぞれ反応させ、抗体結合部位をペロキシダーゼによる組織化学的方法、または、蛍光抗体法にて検出した。これにより、各週齢のヒト胎児皮膚におけるcornified cell envelope構成蛋白(loricrin、small proline-rich proteins、involucrin)の発現の有無、さらに、光顕レベルでの局在部位が明らかになった。
(2)ヒト胎児皮膚におけるcornified cell envelopeの形成過程の超微形態学的観察
胎生8週から24週のヒト胎児皮膚より、電顕用ブロックを作成、超薄切片を作り、それぞれの週齢におけるcornified cell envelopeの形成過程を透過型電顕にて観察した。
(3)先天性掌蹠角皮症患者皮膚におけるcornified cell envelopeの形成の免疫組織学的,および,超微形態学的観察
正常ヒト胎児皮膚におけるcornified cell envelopeの形成過程を角皮症患者皮膚におけるものと比較検討するため,患者本人の同意のもとに先天性掌蹠角皮症患者の皮膚を採取,透過型電顕にて観察した。さらに、cornified cell envelope構成蛋白の局在部位を明らかにするため,抗ヒトloricrin、small proline-rich proteins、involucrin抗体を用いて,蛍光抗体法,および,免疫電顕法により観察した。
