| Research Abstract |
(1) 凍結固定、凍結置換法を用いたPost-embedding免疫電顕法によるcornified cell envelope構成蛋白のヒト胎児皮膚における超微形態学的局在の観察
ヒト胎児皮膚を細切、凍結固定装置を用いて、液体プロパン中に急速に挿入し、凍結固定を行った。この方法によって得られた各週齢の胎児皮膚を超低温に保ったままアセトン置換(凍結置換)、Lowicryl K11Mに包埋、紫外線重合させ免疫電顕用ブロックを作成した。それらより,超薄切片を作成し、抗ヒトloricrin、small proline-rich proteins、involucrin抗体、金コロイド標識2次抗体と反応させたのち、それぞれのcornified cell envelopeの前駆蛋白の超微形態学的局在部位を電顕的に観察した。
(2) 観察結果の総合的評価
ヒト胎児皮膚におけるcornified cell envelope構成蛋白についての、免疫組織化学法、蛍光抗体法、および、Post-embedding免疫電顕法による局在部位の観察結果とcornifed cell envelopeの形成過程の超微形態学的所見の観察の結果を総合し、cornified cell envelope構成蛋白がヒトの発生過程において、いつ、どこに、どのように発現してくるかを厳密に明らかにし,ヒト胎児皮膚におけるcornified cell envelopeの形成部位,および,形成過程を各発生段階毎に明らかにした。
