Research Abstract	モノクローナル抗体を放射性同位元素(RI)で標識して悪性腫瘍のイメージングや内部放射線療法を行う試みが現在まで数多くなされているが、RI標識抗体が血中プールや正常組織に留まり病変と正常組織のRI集積比(腫瘍/正常組織比)が不十分である点が問題であった。解決策のひとつとしてpretargetingの方法が考案された。本研究において検討したのは腫瘍関連抗原とハプテンに特異性のあるBispecific Antibody(BsAb)とRI標識ハプテンを用いる方法である(2ステップ法)。癌胎児性抗原CEAを発現している腫瘍をヌードマウスに皮下移植し継代した。さらに1ステップ法として抗CEA抗体を直接I-125標識した。2ステップ法、1ステップ法それぞれのマウスを経時的に屠殺し放射能の体内分布を調べた。マウスにおける体内分布は2ステップ法では、1ステップ法と同等の腫瘍集積が認められた。2ステップ法の腫瘍/正常組織比は早期から高く、腫瘍血液比は5時間後に6、24時間後に37、48時間後に66、96時間後182と極めて高い値に達した。I-125のデータを基にしてI-131を用いた場合の吸収線量を求めると、腫瘍、肝、腎の吸収線量は2ステップ法で5.50、0.15、0.31、1ステップ法で6.80、0.39、0.69cGy/μCiと計算された。2ステップ法を用いて正常組織の被曝を抑えながら腫瘍に治療量の内部放射線照射を行えることを示しており、放射免疫療法への応用が期待される。
