| Research Abstract |
DNAマーカーによる染色体欠失(LOH)判定を手術時の切除乳癌の遺伝子診断予後の判定に用いるために、乳癌根治術後、死の転帰をとった死亡群57例と術後5年以上の生存群95例との間で24カ所の染色体領域に存在する33個のDNA多形性マーカー座におけるLOHの頻度を比較検討した。さらにプロスペクティブ研究として術後5年以上経過した264症例についてLOHの有無と生存率との関連を比較検討したところ、レトロスペクティブ研究では24領域のうち7領域において、生存群に比べ死亡群において有意に高頻度なLOHを検出した。またプロスペクティブ研究では4領域においてLOH(+)群において不良な予後がKaplan-Meier解析およびLog-lank検定において認められた。また、これらの複数の領域においてともにLOHを示した症例では、5年後の乳癌再発死亡の相対危険率は、LOH(-)群に較べ、8〜10倍に上昇することが明らかとなった。
