ヒト血管内皮細胞の細胞増殖にともなう電気生理的性質

Research Abstract

ヒト臍帯静脈内皮細胞に存在するカルシウム依存性カリウムチャネルの遺伝子解析を行い、以下の結果を得た。なおとの成果の一部はThe International Symposium on Endothelial Function(大阪、1998年10月24日)にて発表済みである。
現在報告されているカルシウム依存性カリウムチャネルの塩基配列を参考にして、大(BKチャネル)および小コンダクタンスカルシウム依存性カリウムチャネル(SKチャネル)のプローブを作成した。継代培養(1代から3代まで)したヒト臍帯静脈内皮由来の血管内皮細胞からメッセンジャーRNAを抽出し、このプローブを用いてRT-PCR法により遺伝子増幅を行い、ヒト臍帯飾脈内皮細胞に発現しているカルシウム依存性カリウムチャネルの遺伝子タイプを決定した。
その結果、BKチャネルはヒト臍帯静脈内皮細胞にほとんど発現していないことが判明した。SKチャネルに関しては現在解析中である。
この結果は、電気生理学的手法を用いてこれまでに得た結果の一部を指示するものであった
