| Research Abstract |
OLETFラットにおけるインスリン非依存性糖尿宿原因遺伝子をクローニングするためにPCR-directed subtraction cloning法を実行した。10週令のOLETF及び対照ラットであるLETOの膵臓より調整されたcDNAをドライバーもしくはトレイサーのいずれかに設定し、過剰ドライバーcDNAとトレイサーcDNA間のハイブリダイゼーション及びドライバーcDNA鎖の除去というサブトラクションを3回繰り返すことでトレイサーcDNA中にのみ存在するcDNAを選抜した。この選抜cDNAPCR産物をプローブとして用いてドライバーcDNA及びトレイサーcDNAを転写したサザンブロットにハイブリダイゼーションさせたところ、ドライバーcDNA及びコントロールの膵RNAのノーザン解析を行ったところ、トレイサーが由来するRNAにより強くハイブリダイゼーションシグナルが観察されると判明した。これらの結果は今回のサブトラクション実験により、OLETFラットの膵臓でコントロールに比べ発現が上昇あるいは減少しているcDNA群が調整されたことを示している。さらに、これらの選抜cDNAPCR産物はpBluescriptベクターにサブクローニングされ、外来インサートDNAとして選抜PCR産物を含むプラスミドが〜100個単離された。これら選抜PCR産物の中に、OLETFラットで生じるインスリン非依存性糖尿病の原因遺伝子となるものもしくはその原因遺伝子の下流で機能を発揮する二次的遺伝子が含まれると考えられる。
