Research Abstract |
1. Immunolight- and immunoelectron-microscopic observation of the localization of NOSs in the normal rat TMJ : Six-week-old male Wistar rats were anesthetized and perfused transcardically with fixative, and TMJs were dissected out. After decalcification, frozen sections were obtained and submitted to immunolight- and immunoelectron-microscopic observation. ABC method with antibodies for eNOS or iNOS was utilized. Immunoreactivity for eNOS and iNOS was found in the synovial lining cells in the anterior and posterior portions of the synovium facing the upper compartment of the TMJ cavity, and in the anterior and lateral portions of the synovium facing of the lower compartment. eNOS was also detected in the endothelial cells of blood vessels close to the synovium. Immunoelectron-microscopic observation revealed that immunoreactivity for eNOS and iNOS was mainly found in the type A cells of the synovial lining cells. In these type A cells, immunoreactive products for eNOS and iNOS were distributed mainly in the cytoplasm, and in the vesicular and vacuolar structures. 2. Immunolight-microscopic observation of the localization of NOSs in the rat TMJ with adjuvant-induced arthritis : Arthritis was induced by the injection of the adjuvant to 7-week-old male Lewis rats. immunolight-microscopic observation for eNOS and iNOS was performed in the same manner as the above mentioned method. The localization of immunoreactivity for eNOS and iNOS in the synovial lining cells of the inflamed TMJ was similar to that of the normal joint. Immunoreactivity for eNOS or iNOS was found in proliferated synovial lining cells facing the upper or lower joint compartment. The strength of immunoreactivity for eNOS was, however, stronger than that for iNOS.Although no clear difference was found in the strength of immunoreactivity for eNOS in the synovial lining cells between the normal rat and the arthritis rat, immunoreactivity for iNOS was reduced in the arthritis rat.
