| Research Abstract |
1.胃癌集積家系の調査、検討、および家系構成者よりのDNA抽出
すでに我々が集め得た7家系の胃癌集積家系の発症年齢や他の構成員の情報の検討、および家系がリンケージ解析に十分な大きさを持つかの点につき検討を行った。また新たな家系の発見および同様の調査を行い、解析に適当であると考えられる家系内において、出来るだけ多くの胃癌患者および健常者の組織(主にリンパ球)よりDNAを抽出した。
2.RDA法によるDNAの解析
RDA法にはBam HI,Hind III,Bgl IIの3種の制限酵素を用いた。同一家系内での胃癌患者と健常者DNAをこれらの制限酵素で切断した後、各々PCR法にて増幅したのちRDA法によりサブトラクションを行った。この方法により、胃癌患者と健常者でDNA多型を示す多数のマーカーの検出を行った。
