宿主拒絶反応に及ぼすヘルペスウイルス感染の影響:マウス臓器移植モデルによる解析

Research Abstract

1,マウスサイトメガロウイルス(MCMV)抗体測定系
MCMV感染MRC-5 cellを12穴スライドグラス上にアセトン固定、間接蛍光抗体法でMCMV IgG抗体を測定する系を確立中。BALB/cマウス腹腔内にSmith株HCMVを接種し、急性期、回復期血清を採取し抗体アッセイ系の感度、特異性を検討中。
2,MCMV分離、培養
培養繊維芽細胞を使い、ウイルス分離ならびに培養を実施。高力価のウイルス液が得られることを確認。
3,MCMVゲノム検索法
MCMV immediate early 1 gene内にプライマーを設定、1701-2400の領域を増幅する。PCRの設定は過去の報告に従い決定した。陽性コントロールを用いた検討で、特異的な遺伝子増幅を確認した。
4,MCMV感染系の確立
BALB/c×C3Hマウスに、2で整節したHCMVを経鼻接種。感染後、経時的にマウスより採血し抗体測定、ウイルスゲノム検索を実施中。
5,今後の予定
4,に述べたMCMV感染系が確立されたら、(1)ドナーマウス感染無し×レシピエントマウス感染無し(2)ドナーマウス感染有り×レシピエントマウス感染無し、(3)ドナーマウス感染無し×レシピエントマウス感染有り、(4)ドナーマウス感染有り×レシピエントマウス感染有りの各系列を作成。それぞれの組み合わせについて肝移植を実施。術後拒絶反応、ウイルス感染動態を解析する。
