CDR-humanized CC49遺伝子の作製
CDR-grafted humanized CC49ではframeworkにlight chainはLENを、heavy chainは21/28′CLを用いており、CDRもそれぞれこれらの同じヒト抗体のCDRと入れ換えることとした。homologyの極めて高いlight chainはsite directed mutagenesisPCRを行いc-DNAを得た.またhomologyの比較的低いheavy chainはDaughertyらの方法(Nucl Acids Res,1991)でPCRを行いc-DNAを得た。これらのPCR productsをTA vectorにcloningし,sequencingを行い正しい塩基配列であることを確認した。light chainはヒトKappa鎖定常領域と,またheavy chainはヒトIgG1の定常領域と結合させ、completeのvariant humanized CC49の遺伝子を作製した(L-CDR 1,L-CDR 2,L-CDR 1,2,L-CDR 3,H-CDR 1,H-CDR 2,H-CDR 3)。
Baculovirus expression vectorの作製とin vitroにおける抗体発現
それぞれのvariantsのheavy chainとlight chainの遺伝子をbaculovirus dual expression vector(pAcUW51)にcloningしWild type virus DNAとともにSf9 cellsにtransfectした。培養上清の蛋白発現をELISAで確認しながらplaque assayにてvirus purificationを行った。その後virus amplificationを行い、再度plaque assayにてtitrationを行い,このvirus液をMOI5にてSf9 cellに感染させ、3日間培養し培養上清を回収した。DE52カラムおよびprotein Gカラムにて7種類の抗体を精製した(0.85〜7.5mgの充分量の抗体を得た)。それぞれの抗体をSDS pageおよびWestern blottingにて純度を確認した。
Affinity competition RIA assay
Controlのhumanized CC49を^<125>Iでlabelし,^<125>I-labeled humanized CC49をcompetitorとしてcompetition RIA assayの準備中である。
