Research Abstract

We visited the counterpart in Semipalatinsk and received informations of leukemia as well as the samples. We reviewed the medical records and smears for 31 cases with leukemia and myelodysplastic syndrome (MDS). Eighteen cases of them were considered to be exposed to radiation associated with nuclear tests, based on that the nuclear tests in air were performed between 1949 and 1963. These cases were studied morphologically and molecular biologically. The 7 were males and 11 females, and 8 were Kazachcians, 8 Ruccians and 2 Germans. The age at diagnosis was between 37 and 78. Six acute myelogenous leukemia (AML) M1, 1 AML M2, 2 AML M6, 1 unclassified AML, 2 acute lymphoblastic leukemia (ALL), 2 unclassified acute leukemia, 1 chronic myelogenous leukemia, 1 MDS-RA, 1 MDS-RAEB and 2 MDS-RAEBt were included. Two leukemia-related genes, p53 and AML1, were analysed by PCR-SSCP and sequencing. Among 1 8 cases examined, a p53 point mutation was revealed at exon 6 of codon 195 ATC(Ile)→ACC(Thr) in one unclassifiable acute leukemia case. AML1 point mutation at runt domain (exon 5) was found in one MDS-RA case. AML1 gene is suggested to be involved in radiation-related MDS and AML.Therefore, more cases should be analysed to see whether this mutation is frequent in this region.
