Research Abstract	本研究は、大量の水分泌と開口分泌を観察できる唾液腺に対象を限定、小胞輸送により運ばれた分泌顆粒が細胞膜に融合し開口放出を起こすプロセスについて共同研究者の各実験サイトで分子生理的、形態学的に水電解質輸送と開口分泌を計測、水輸送・開口分泌両システム間の協調機構を検討することを目的とする。平成10年度、 A. 分泌経過チーム:酵素開口分泌(漿液分泌)のモデルとして、耳下腺の血管潅流・単離細胞標本を作製し、カルバコール(ムスカリニック刺激)イソプロテレノール(βアドレナジック刺激)について水電解質分泌・アミラーゼ分泌・酸素消費の時間経過を測定、水分泌と開口分泌が、前者は主として細胞内Ca信号によりコントロールされ、後者はCaとcyclic AMP両者の組み合わせで制御を受けることが明らかになった。 B. 形態チームは、レーザー・走査電子顕微鏡を用い、Aのプロトコルで、耳下腺の血管潅流・単離細胞標本の「分泌顆粒、小胞輸送、膜融合、開口放出、膜の回収」の時間経過に沿って水経路の形態の時間的変化とアミラーゼ開口分泌の微小形態の変化を観察した。その結果カルバコール刺激では大きな形態変化は無く、細胞間小管の拡大がみられるのみであった、一方開口分泌像はカルバコールとイドプロテレノール両者の刺激で多く観察され、貯蔵された分泌顆粒もその数を減少させた。 C. 開口放出関連蛋白チームは、耳下腺分泌顆粒膜の安定化を検討した。また、ターゲットの細胞膜における局在を、形態グループと協力して実施するために、新しいSEM法の開発に着手した。 D. 分泌物質の運動性グループは、本年は、水分泌経路のサイズの測定のため研究協力者ヒル博士(ケンブリッジ大と共同実験を行い標識デキストランの腺房上皮通過を測定した。本年度、アミラーゼ分泌増加と対応した酸素消費増加分を蛋白分泌に必要なエネルギー要求として測定できた。
