Research Abstract	第11染色体q23領域に存在するヒト肺非小細胞がん(NSCLC)抑制遺伝子を単離する目的で、酵母人工染色体(YAC)クローンをヒトNSCLC細胞A549に導入し、ヌードマウス皮下の腫瘍原性の抑制を指標として遺伝子座位を100kbに退局化し、新規候補遺伝子TSLC1/IGSF4cDNAを単離した。この遺伝子は一回膜貫通ドメインをもつ糖蛋白質をコードし免疫globulin superfamily、特に神経細胞接着因子(NCAM)と構造類似性を示す。この遺伝子に関して、以下の知見を得た。 (1)検索したすべての組織で発現を認めたが、A549を含むNSCLC培養細胞12例中8例ではmRNAの欠如、ないし著明低下を認めた。またヒトNSCLC腫瘍で高頻度にLOHを示した。 (2)全長cDNAをNSCLC細胞A549に導入すると、正常肺組織相当の発現量で腫瘍原性が抑制された。 (3)N端のシグナルペプチドを欠如し、細胞膜に局在しない分子は腫瘍抑制活性を示さなかった。 (4)遺伝子産物が細胞膜、並びに核周辺の膜構造に局在し、糖鎖附加を受けることを実験的に示した。以上の知見により、TSLC1/GSF4は新規肺非小細胞がん抑制遺伝子であると結論した。さらに肺がん以外の様々なヒトのがんにも関与することを示唆する知見が得られている。
