巨大血管腫特異的な遺伝子群の腫瘍新生血管における機能

Research Abstract	癌の血管新生に特異的な分子標的を単離することは、癌の病態を把握するだけでなく新しい診断と治療のマーカーとして提供する上で極めて重要である。そこで本研究では肝臓に生じた増殖が早く、手術後くり返し再発する血管腫を対象に増殖が遅い血管腫mRNA間でDifferential Display(DD)を行ない特異的な遺伝子を単離することにした。さらに肝臓に生じた肝細胞癌(HCC)と胆管癌(CCC)の血管新生の高低を左右する因子についての検討を進めることにした。その結果、 1. DD法で得られたクローンのうち増殖の早い血管腫に発現が高い5クローンを単離した。5クローンについての部分塩基配列から新しい遺伝子であることが示唆された。 2. 細小血管密度の高いHCC症例では、非癌部位に較べて癌部位で促進因子、血管内皮増殖因子(VEGF)の発現が高く阻害因子トロンボスポンジン1(TSP-1)は低かった。他方、血管密度が低いCCCではVEGFの発現は低く、TSP-1の発現は高かった。以上の結果をもとに、血管内皮に発現する新しい遺伝子の単離と構造、機能の解析さらに血管新生阻害因子TSP-1の発現の機構について現在仕事を進めている。
